Solid Phase Assays

 

Multiwell Plate Assays

There are some multiwell plate solid phase binding assays, which appropriate Biotech has been contracting to bioanalytical CROs for preclinical and clinical drug development of investigational new nucleic acid therapeutics:

  • sandwich hybridisation ligand binding assays for the ultra-sensitive quantification of siRNA, aptamer, or microRNA therapeutics in biological samples collected in concomitant pharmacokinetic studies,
  • antibody tools for nucleic acid binding assays to combine sandwich hybridisation techniques with the advantages of (monoclonal) antibodies for increased selectivity,
  • branched DNA hybridisation binding assays as an alternative to liquid phase Real-Time RT-qPCR for transcript quantification, which often redundantizes the need for RNA extraction,
  • antibody detection binding assays (ADA) to detect any innate immune response to administered nucleic acid therapeutics,
  • plasma protein ligand binding assays, which help to better understand the (unspecific) binding of plasma proteins to the nucleic acid drug both in vitro and in vivo, though these kind of assays are usually performed on filters,
  • enzyme-linked immunospot (ELISpot) ligand binding assays are designed to detect immune response (like cytokine excretion) on a single cell level, though these kind of assays are usually performed on membranes at the bottom of multiwell plates,
  • immuno qPCR ligand binding assays are designed to combine the specificity of ligand-binding antibodies with the sensitivity of the polymerase chain reaction (PCR). The bead assay format may improve ligand binding capacity through the almost globular surface of the magnetic particles,

On-Bead Ligand Binding Assays

There are some solid phase binding assays involving microspheres or magnetic beads, which appropriate Biotech has been contracting to bioanalytical CROs for preclinical and clinical drug development:

  • Luminex® ligand binding assays for the ultra-sensitive detection of proteins, peptides, and nucleic acids,
  • Systematic evolution of ligands by exponential enrichment (SELEX) ligand binding assays for the ultra-sensitive selection of nucleic acid ligands specific for peptides or proteins,
  • Immuno qPCR ligand binding assays for the ultra-sensitive quantification of  peptide or protein ligands, an underestimated assay alternative to ELISAs. The microwell plate format allows for ligand immobilization and qPCR assay in the same plate,
